This article explains the roles of all the various game settings. They were split into two categories for clarity.
There are 4 difficulties available to pick on the creation screen when naming your company, these are:
For casual gameplay without worry.
The standard difficulty. Everything is optimized to play well on this difficulty.
A challenge for less-experienced players and probably the base difficulty for experienced players.
Very Hard (V. Hard)
A challenge for the vast majority, needing constant attention and possibly micromanagement, while attempting to stay fair.

You can customize your playthrough with settings accessible by clicking on the little cog in the bottom-right corner of the creation window.
Lets you select the campaign you want to play through from a drop-down list.
Checked by default. If unchecked, removes the possibility of losing the game by having negative balance for 120 consecutive days. However, it's still possible to get "stuck" by running out of money and taking out all loans in the bank. Decreases your high score by ~13%.
Checked by default. If unchecked, removes any traces of competitors, such as news and their products on the market. Has next to no impact on gameplay. Doesn't affect the high score.
Unchecked by default. If checked, gives you infinite money. It's impossible to run out of it, unless the console is used. Also called sandbox mode. Reduces your high score by 98%.
